The Science & Mechanism

Secretory IgA (sIgA) is a dimeric immunoglobulin produced by plasma cells in the intestinal lamina propria, where two IgA monomers are joined by a J chain. To reach the lumen, it binds the polymeric immunoglobulin receptor (pIgR) on the basolateral surface of epithelial cells and is transcytosed across; cleavage of pIgR releases the antibody with a bound secretory component that resists degradation by intestinal proteases and helps anchor it within the mucus. At the surface, sIgA works largely through non-inflammatory mechanisms. In immune exclusion, it agglutinates bacteria and cross-links antigens to trap them in mucus, limiting contact with the epithelium, while also neutralizing toxins and viruses. Research describes how sIgA coats commensal microbes and thereby helps shape microbiome composition and spatial organization, promoting tolerance to the resident community. Its production is influenced by mucosal signaling, including T cell help, cytokines such as TGF-beta, and microbial cues, and it is a major arm of gut-associated lymphoid tissue.
