How It Works

Vitamin K2 is a fat-soluble vitamin closely related to the K1 found in leafy greens, but its forms—called menaquinones—behave differently in the body. Its central job is to switch on certain proteins that decide where calcium ends up: it helps activate proteins that bind calcium into bone and others that keep calcium from settling into artery walls and soft tissue. In everyday terms, K2 acts like a traffic director for calcium. Because it works hand-in-hand with vitamin D, which raises calcium availability, the two are often considered together. The Science & Mechanism

Vitamin K2 functions as an essential cofactor for the enzyme gamma-glutamyl carboxylase, which converts specific glutamate residues on "Gla proteins" into gamma-carboxyglutamate. This carboxylation step gives these proteins their calcium-binding capacity. Two are central to K2 research: osteocalcin, produced by osteoblasts, which in its carboxylated form helps incorporate calcium into the bone matrix; and matrix Gla protein (MGP), which in its active form inhibits the deposition of calcium in arterial walls and other soft tissues. Uncarboxylated forms of these proteins are inactive and are used in research as markers of vitamin K status. During carboxylation, vitamin K is oxidized to its epoxide form and then regenerated by the enzyme vitamin K epoxide reductase in what is known as the vitamin K cycle. K2 occurs as several menaquinones (MK-n) distinguished by side-chain length; the shorter MK-4 and longer, longer-circulating MK-7 are the most studied. Compared with K1, menaquinones tend to have greater bioavailability to extrahepatic tissues such as bone and vasculature, which is why research explores K2 specifically for calcium distribution beyond the liver.
