The Science & Mechanism
Niacin is the collective name for nicotinic acid and nicotinamide, both precursors to nicotinamide adenine dinucleotide (NAD+) and its phosphorylated form NADP+. These coenzymes are central electron carriers: NAD+/NADH shuttle reducing equivalents through glycolysis, the citric acid cycle, and oxidative phosphorylation, while NADP+/NADPH support reductive biosynthesis and antioxidant defense via glutathione regeneration. Beyond its coenzyme role, NAD+ is a required substrate for enzymes that consume it, including sirtuins (SIRT1-7), which influence mitochondrial function and stress responses, and PARPs, which participate in DNA-damage repair. Nicotinic acid also acts as a ligand for the G-protein-coupled receptor HCA2 (GPR109A) on adipocytes and immune cells; engagement of this receptor is associated with reduced lipolysis and is the basis of research into its effects on circulating lipids, as well as the characteristic flushing response mediated by prostaglandins. The body synthesizes some NAD+ de novo from tryptophan and salvages it through pathways using nicotinamide and related precursors, making dietary B3 an important contributor to overall NAD+ availability.
