Vitamin B6 is an umbrella term for six related compounds, including pyridoxine, pyridoxal, and pyridoxamine and their phosphorylated forms. The metabolically active coenzyme is pyridoxal 5'-phosphate (PLP), which serves as a cofactor for more than 140 distinct enzyme activities, a large share of all known catalytic reactions that require a coenzyme. PLP is central to amino-acid metabolism, participating in transamination, decarboxylation, and deamination reactions that reshape the body's nitrogen-containing building blocks. Through decarboxylase enzymes, PLP is required for the synthesis of several neurotransmitters, including serotonin from tryptophan, and dopamine, norepinephrine, and GABA. It contributes to the first committed step of heme synthesis via ALA synthase and supports glycogen breakdown through glycogen phosphorylase. PLP also functions within one-carbon metabolism: as a cofactor for cystathionine beta-synthase and cystathionine gamma-lyase in the transsulfuration pathway, it is associated with the clearance of homocysteine. Research further describes B6's involvement in immune signaling and its activity as a quencher of certain reactive oxygen species, reflecting the breadth of physiology that depends on adequate status.
