Vitamin B12 is one of the eight B vitamins, distinguished by the trace mineral cobalt at the center of its structure. The Science & Mechanism
Vitamin B12 functions as a cofactor for two enzymes central to human metabolism. In the cytoplasm, methylcobalamin supports methionine synthase, which converts homocysteine to methionine and regenerates tetrahydrofolate. This reaction links B12 to folate metabolism and to the methylation cycle that generates S-adenosylmethionine, a universal methyl donor for DNA, RNA, proteins, and neurotransmitters. In the mitochondria, adenosylcobalamin supports methylmalonyl-CoA mutase, which converts methylmalonyl-CoA to succinyl-CoA, feeding fatty acid and amino acid breakdown into the citric acid cycle for energy production. When B12 is insufficient, homocysteine and methylmalonic acid accumulate, and impaired methylation slows DNA synthesis in dividing cells such as bone marrow precursors, producing the enlarged red cells of megaloblastic anemia. B12 is also studied for its role in maintaining the myelin sheath, and inadequate methylation is associated with neurological changes. Dietary B12 requires stomach acid, pepsin, and intrinsic factor secreted by gastric parietal cells for absorption in the terminal ileum, so research describes how gastrointestinal conditions, certain medications, and normal aging can reduce uptake over time.
