The Science & Mechanism
Iodine is an essential trace element whose primary biological role is as a structural component of the thyroid hormones thyroxine (T4) and triiodothyronine (T3). Circulating iodide is actively concentrated in thyroid follicular cells by the sodium-iodide symporter (NIS), then oxidized and organified onto tyrosine residues of thyroglobulin by thyroid peroxidase, a reaction that also requires hydrogen peroxide. This produces mono- and diiodotyrosines that couple to form T4 and, to a lesser degree, T3. T4 is the main secreted form and is later deiodinated in peripheral tissues by selenium-dependent deiodinases to the more active T3, which binds nuclear thyroid hormone receptors to regulate gene transcription. Through this pathway, thyroid hormones influence basal metabolic rate, thermogenesis, protein synthesis, and neurodevelopment. Iodine status also interacts with selenium and iron, cofactors needed for hormone synthesis and activation. Research describes the thyroid's autoregulation of iodine uptake, and both insufficient and excessive intake are studied in relation to thyroid function, making balanced status the focus of physiologic interest.
