Mechanistically, withanolides are studied for GABA-mimetic activity, meaning they appear to interact with GABAergic pathways that quiet excitatory signaling in the brain, an action that may help explain observed effects on relaxation and sleep. Preclinical work also explores antioxidant and anti-inflammatory signaling, including modulation of Nrf2-linked defenses and NF-kB-driven inflammatory pathways, as well as support for mitochondrial function.
