KPV is a short peptide built from three amino acids that mirrors the active tail of a natural anti-inflammatory hormone. Once inside cells, it is thought to quiet the molecular switches that drive inflammation, which may help tissue settle and recover. The Science & Mechanism
KPV is the C-terminal tripeptide fragment (Lysine-Proline-Valine) of alpha-melanocyte-stimulating hormone (alpha-MSH), and it appears to retain much of that parent hormone's anti-inflammatory activity in a smaller, more stable form. Research suggests KPV acts intracellularly to downregulate pro-inflammatory signaling cascades, notably by interfering with NF-kB and MAPK pathway activation and reducing downstream production of inflammatory cytokines such as TNF-alpha, IL-1beta, and IL-6. In epithelial and immune cells it is thought to be taken up in part via the peptide transporter PepT1, which may concentrate its activity in the gut lining. Unlike full-length alpha-MSH, KPV appears to exert these effects with minimal melanocortin-receptor-mediated pigmentation activity, which is one reason it is studied as a focused anti-inflammatory peptide.
