The Science & Mechanism

BPC-157 is a synthetic 15-amino-acid peptide whose sequence corresponds to a partial fragment of a protective protein identified in gastric juice. A central mechanism studied in preclinical models is promotion of angiogenesis: it has been shown to upregulate VEGF and activate the VEGFR2 receptor, engaging downstream PI3K/Akt and endothelial nitric oxide synthase (eNOS) signaling, alongside a VEGF-independent Src - Caveolin-1 - eNOS pathway. Both routes converge on nitric oxide production, which appears to be modulated in a context-dependent manner that may support healthy vascular tone and microcirculation at sites of injury. Research also describes upregulation of the early growth response gene-1 (Egr-1) and its co-repressor, increased expression of growth factors such as FGF and PDGF, and influence on the FAK - paxillin pathway associated with fibroblast migration. Together these actions are studied for their role in the formation of new blood vessels and organized collagen during repair.
