The Science & Mechanism
IGF-1 LR3 (Long R3 IGF-1) is a recombinant analog of insulin-like growth factor 1 with two structural changes: a 13-amino-acid extension on the N-terminus and a substitution of arginine for glutamic acid at position 3. Together these markedly lower its affinity for the IGF-binding proteins (IGFBPs) that normally sequester circulating IGF-1, leaving a larger free fraction and giving it a substantially longer functional half-life. It acts primarily at the IGF-1 receptor, a tyrosine kinase that activates the PI3K/Akt and MAPK pathways governing protein synthesis, cell proliferation, differentiation, and survival, while also stimulating cellular glucose and amino-acid uptake. Native IGF-1 mediates many of growth hormone's downstream anabolic effects and is produced largely in the liver and in local tissues. Because the LR3 form evades normal IGFBP regulation, research describes more sustained receptor signaling than endogenous IGF-1 would produce. Most existing data derive from laboratory and cell-culture contexts, including its widespread use as a cell-culture growth supplement; rigorous human clinical characterization of the LR3 analog remains limited.
