The Science & Mechanism
FGL is a synthetic peptide derived from the second fibronectin type III module of the neural cell adhesion molecule (NCAM), reproducing the FG loop region that NCAM normally uses to contact the fibroblast growth factor receptor (FGFR). By mimicking this contact, FGL is thought to act as an FGFR agonist, triggering receptor dimerization and autophosphorylation. This is believed to activate downstream cascades associated with neuronal survival and plasticity, including the MAPK/ERK and PI3K/Akt pathways and CREB-mediated gene transcription. Preclinical research suggests that engaging this axis may promote synaptogenesis, support long-term potentiation, and increase presynaptic markers in regions such as the hippocampus. Investigators have also explored anti-inflammatory and neuroprotective actions, including modulation of microglial activity and protection of neurons under metabolic or excitotoxic stress.
