DSIP is a small endogenous nonapeptide (Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu) first identified in the cerebral venous blood of sleeping rabbits. Rather than acting as a classical sedative, it is thought to function as a neuromodulator that interacts with multiple central systems, and research has explored its influence on the balance of excitatory and inhibitory neurotransmission, including glutamatergic and GABAergic tone. Studies suggest it may interact with the hypothalamic-pituitary-adrenal (HPA) axis and related corticotropin and cortisol signaling, and it has been investigated for possible interactions with opioidergic and serotonergic pathways thought to shift sleep architecture toward slow-wave (delta) activity. It has also been studied in the context of circadian and thermoregulatory signaling, which is why its effects are generally described as rhythm-modulating rather than directly hypnotic. Much of this mechanistic understanding comes from preclinical and early research rather than established clinical consensus.
