The Science & Mechanism
Sleep is orchestrated by the circadian system and homeostatic sleep pressure, and it directly shapes immune and inflammatory signaling. During slow-wave sleep, activity of the hypothalamic-pituitary-adrenal axis and sympathetic nervous system declines, lowering cortisol and catecholamines that otherwise modulate immune cells. Research describes sleep loss and fragmentation as activating nuclear factor kappa B (NF-kB), a master transcription factor for inflammatory genes, with associated rises in interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-alpha), and C-reactive protein (CRP) in many studies. Sleep also entrains the diurnal rhythm of these cytokines and of immune-cell trafficking, so mistimed or curtailed sleep can flatten or shift those rhythms. The hormone melatonin, released as darkness falls, is studied both as a circadian signal and as an antioxidant that interacts with inflammatory pathways. Deep sleep supports glymphatic clearance of metabolic waste in the brain and healthy growth-hormone pulses tied to tissue repair. Experimental sleep restriction in humans is associated with increased inflammatory markers, altered natural killer-cell activity, and blunted responses in some vaccination studies, while consistent, sufficient sleep is associated with more balanced immune function.
