How It Works

Melatonin is a hormone produced mainly by the pineal gland, a small structure deep in the brain, and it is released chiefly at night. In everyday terms, it acts as the body's darkness signal: as evening light fades, melatonin rises and quietly tells the brain and body that night has arrived and it is time to wind down. Rather than acting like a sedative that forces sleep, it works more like a timekeeper that opens the biological window for rest and helps align your internal clock with the day-night cycle. Its natural rise and fall each 24 hours is one of the clearest markers of circadian rhythm, and evening light, especially blue-enriched screen light, can blunt or delay it. The Science & Mechanism

Melatonin (N-acetyl-5-methoxytryptamine) is synthesized from the amino acid tryptophan by way of serotonin, which is converted to melatonin in the pineal gland through the rate-limiting enzyme arylalkylamine N-acetyltransferase (AANAT) and then a methyltransferase. Its production is governed by the suprachiasmatic nucleus (SCN), the brain's master clock in the hypothalamus, which receives direct light input from specialized retinal cells containing melanopsin. Light suppresses melatonin release, so secretion is low by day, rises in the evening, peaks overnight, and falls before waking, producing a robust circadian curve. Once released, melatonin acts largely through two high-affinity G-protein-coupled receptors, MT1 and MT2, which are densely expressed in the SCN itself. Through these receptors melatonin feeds back on the clock to reinforce nighttime signaling and to shift circadian phase, an action captured by the phase-response curve. Research also describes melatonin as a lipophilic molecule that crosses membranes readily and functions as a direct and indirect antioxidant. Endogenous levels are highest in childhood and tend to decline with age, and its timing, more than its absolute amount, is what research emphasizes for circadian alignment.
