Circulating GH binds the growth hormone receptor, a class I cytokine receptor, triggering JAK2-STAT5 signaling that drives transcription of target genes. Many of its anabolic effects are indirect, mediated by insulin-like growth factor 1 (IGF-1), produced mainly by the liver, which promotes cell proliferation, protein synthesis, and tissue growth; IGF-1 in turn feeds back to suppress GH release, closing a regulatory loop. GH also acts directly, exerting lipolytic effects that mobilize fat, and it can antagonize insulin, influencing glucose handling.
