They function largely as prohormones: through the sequential action of enzymes such as 3-beta-hydroxysteroid dehydrogenase, 17-beta-hydroxysteroid dehydrogenase, and aromatase, DHEA is converted in peripheral tissues into androgens and estrogens, allowing local tissue-specific hormone production known as intracrinology. Beyond serving as a precursor, DHEA is studied for direct and indirect actions at androgen and estrogen receptors and as a neurosteroid that modulates GABA-A and NMDA receptor signaling in the brain. Research also explores its relationship to the hypothalamic-pituitary-adrenal axis, where DHEA is often discussed alongside cortisol and framed as a counterbalancing signal, with the cortisol-to-DHEA ratio examined as a marker of stress physiology.
