How It Works

The Science & Mechanism

MOTS-c is a 16-amino-acid peptide encoded within the 12S rRNA region of mitochondrial DNA, making it one of the first identified mitochondrial-derived peptides. It acts largely through the folate cycle - AICAR - AMPK pathway: by modulating the folate cycle and its linked de novo purine synthesis, it promotes accumulation of AICAR, an activator of AMP-activated protein kinase (AMPK), a central metabolic energy sensor. AMPK activation is associated with GLUT4-mediated glucose uptake into skeletal muscle and enhanced mitochondrial fuel utilization, paralleling some of the metabolic signaling seen with physical exercise. Under metabolic stress, MOTS-c has also been observed to translocate from the mitochondria into the cell nucleus, where it appears to help regulate stress-adaptation genes, including antioxidant response element (ARE) - linked pathways. Together, these actions are studied for a role in energy homeostasis, glucose handling, and cellular stress resilience.
