The Science & Mechanism
Melanotan II is a synthetic cyclic heptapeptide analog of alpha-melanocyte-stimulating hormone (alpha-MSH), with the sequence Ac-Nle-c[Asp-His-D-Phe-Arg-Trp-Lys]-NH2. Its cyclic structure resists enzymatic breakdown, giving it greater stability and potency than the native hormone. It acts as a non-selective agonist across the melanocortin receptor family, binding MC1R, MC3R, MC4R, and MC5R. Its best-characterized action is at MC1R on melanocytes: receptor activation raises intracellular cyclic AMP, which activates protein kinase A and the CREB transcription factor, in turn upregulating microphthalmia-associated transcription factor (MITF), the master regulator of melanogenesis. This drives synthesis of eumelanin, the brown-black pigment that absorbs UV photons, scavenges free radicals, and physically shields melanocyte DNA. It was originally developed at the University of Arizona in the 1980s as a candidate photoprotective agent intended to build protective pigment in people with extreme sun sensitivity. Because it also engages central MC3R and MC4R, research describes additional effects on appetite and sexual response, which is why its non-selective activity is closely considered. Its shorter, more selective cousin afamelanotide (Melanotan I) is a linear MC1R-preferring analog that reached European approval for a rare photosensitivity disorder.
