The Science & Mechanism

LL-37 is the active 37-amino-acid peptide cleaved from the C-terminal end of hCAP18, the single cathelicidin protein encoded by the human CAMP gene; its name reflects the two leucine residues at its start. As a cationic, amphipathic alpha-helical peptide, it is studied for a direct antimicrobial mechanism in which its positive charge draws it to the negatively charged membranes of bacteria, fungi, and enveloped viruses, where it inserts and disrupts membrane integrity. Beyond this, research describes LL-37 as an immunomodulatory host-defense peptide that signals through the formyl peptide receptor-like 1 (FPRL1/FPR2) and can transactivate the epidermal growth factor receptor (EGFR). Through these pathways it is associated with chemotaxis of neutrophils, monocytes, and T cells, promotion of angiogenesis and wound re-epithelialization, and neutralization of bacterial lipopolysaccharide (LPS) to temper excessive endotoxin-driven inflammation. It is also studied for context-dependent effects on inflammatory signaling and for supporting epithelial barrier function at the skin and gut. Its expression is influenced by vitamin D signaling, which research explores as a link between vitamin D status and innate immunity.
