GHRP-6 is a synthetic hexapeptide (His-D-Trp-Ala-Trp-D-Phe-Lys-NH2) and a first-generation growth hormone releasing peptide. It acts as an agonist of the growth hormone secretagogue receptor (GHS-R1a), a G protein-coupled receptor expressed on somatotroph cells of the anterior pituitary and in hypothalamic centers that regulate appetite and energy balance. Receptor activation engages the phospholipase C pathway, generating IP3 and diacylglycerol, mobilizing intracellular calcium, and driving the pulsatile release of endogenous growth hormone. GHRP-6 also promotes GH secretion indirectly by stimulating GHRH-producing neurons in the hypothalamic arcuate nucleus and by blunting the inhibitory tone of somatostatin, the hormone that normally restrains GH output. Because GHS-R1a is the native receptor for ghrelin, the body's principal hunger hormone, GHRP-6 characteristically stimulates appetite, which distinguishes it from more selective secretagogues such as ipamorelin. Historically, GHRP-6 was the pharmacological probe that predicted and led to the discovery of ghrelin and its receptor, making it a landmark research compound. Beyond growth hormone, research also explores its interaction with the CD36 scavenger receptor and possible cytoprotective and anti-inflammatory signaling in various tissues, effects described as at least partly independent of growth hormone release.
