Adiponectin is a peptide hormone, or adipokine, secreted almost exclusively by adipocytes and circulating at high concentrations relative to other hormones. It assembles into low-, medium-, and high-molecular-weight multimers, with the high-molecular-weight form generally considered the most metabolically active. It signals mainly through two receptors, AdipoR1 (abundant in skeletal muscle) and AdipoR2 (abundant in liver), along with T-cadherin in vascular tissue. Receptor engagement activates AMP-activated protein kinase (AMPK) and PPAR-alpha pathways, which promote fatty-acid oxidation, glucose uptake, and reduced hepatic glucose production, effects that research links to improved insulin sensitivity. Adiponectin is also studied for anti-inflammatory and vascular actions, including influence on endothelial function and nitric oxide signaling. Notably, circulating levels are inversely associated with visceral adiposity, insulin resistance, and inflammatory markers, so higher adiponectin generally tracks with a healthier metabolic profile.
